First, the advice.
Always roll-step. That does wonders for everything else.
Now the answer.
Best way to breathe is large frequent breaths through the mouth, with even more quick breaths through the nose whenever possible. Make sure you hold the sousaphone in a way that keeps the weight from being supported my your stomach. Even though it hangs on your shoulder, if the valve tubing is touching your stomach area it will be hard to breathe.
