If the bell isn't too big, use the trunk. If you are concerned about sliding, get one of those nets they sell at pickup truck accessory shops to hold it in place. Trust me if it's held down in your trunk with one of those you'll have more to worry about than your tuba if you get hit hard enough to move that horn around.
Most of my cars won't take my big-belled horns in the trunk, so I personally use the passenger seat, with the harness buckled across the horn. If the seats fold forward and I am carrying more than one horn, use the back seat with the horn laid such that the bell side is facing the back of the back seat, then recline the passenger seat enough to "nest" the horn in place. Again, if this is insufficient you'll have more to worry about than your tuba.
