See a doctor... and he will tell you exactly what you should and should not be doing with that lip of yours.
It sounds like a serious injury and you shouldn't push yourself through practising (and auditioning) before it heals. A tuba player is an athlete just like a football player (for example) - and while, in the playoffs, athletes are used to playing injured, they will routinely take 4 or 5 weeks off to heal and rehabilitate during the regular season. Bottom line: if tuba playing is so important to you, then take care of your chops and rest them until you can blow without pain. Then take it slow on longtones to rebuild.
