You probably want opinions from tubists who have actual experience with this, but in my opinion most mouthpieces have been designed to work with the throat size they have already, and you don't improve the overall playing characteristics by drilling it out. You might make one aspect easier, but at the expense of other characteristics. Somebody went to the trouble of designing the mouthpiece to play as it is...don't destroy it, unless it's worthless as it is.
